Description

Trichodermamide B is a JAK/STAT3 inhibitor. Trichodermamide B exhibits a potent antiproliferative activity with an IC50 value of 0.12 μM against HCT116. Trichodermamide B induces cell apoptosis and has anti-cancer effects[1].

In Vitro

Trichodermamide B (compound 24; 0-5 μM; 72 h) displayed potent activities in inhibition on the proliferation of cancer cells in a concentration-dependent manner, with IC50 values of 4.79, 0.90, 4.15, 0.53, 0.66, and 0.12 μM for A549, DU145, Hela, MDA-MB-231, MCF-7, and HCT116, respectively[1].
Trichodermamide B (0-5 μM; 1-2 weeks) is able to inhibit cell colony formation in A549 and DU145 cell lines at the low concentration of 0.5 μM[1].
Trichodermamide B (0.5-5 μM; 48 h) inhibits the migration of A549 and DU145 cells in a dose-dependent manner[1].
Trichodermamide B (2.5-10 μM; 24 h) induces apoptosis in A549 and DU145 cells as illustrated by flow cytometry[1].
Trichodermamide B (1-10 μM; 0-24 h) blocks JAK/STAT3 signaling in dose- and time-dependent manner[1].

Trichodermamide B (compound 24; 10 or 20 mg/kg; i.p.; daily; for 14 days) displays potent in vivo efficacy with a tumor growth inhibition (TGI) rate of 65% at 20 mg/kg. Trichodermamide B inhibits the STAT3 phosphorylation level and the amount of its downstream proteins CyclinD1 and Bcl-xL in the tumor[1].
