Definition:

Mammalian Protein-Protein Interaction Trap (MAPPIT) is a protein-protein interaction screening method based on the reconstruction of membrane STAT (transcriptional activator) receptors in mammalian cells. The bait protein is fused with a receptor lacking the STAT recruitment site, while the prey protein is fused with a functional STAT recruitment site. In this configuration, the given bait-prey interaction restores a STAT-dependent response, leading to the expression of a reporter gene. This system can not only display protein interactions but also interactions that are independent of modifications but depend on tyrosine phosphorylation.
