Protein Three Hybrid

Definition:

Protein Three Hybrid is based on the classical two-hybrid technique with the addition of a third protein component to detect interactions among the three. In the experiment, the bait protein, prey protein, and the third protein are co-transfected into recombinant yeast. Negative control experiments show that the interaction between bait and prey does not occur unless the third protein is co-transfected. This technique is commonly used to study the assembly and regulatory mechanisms of multi-protein complexes.
