Reverse Two Hybrid

Definition:

Reverse Two Hybrid is used to study specific protein-protein interactions, especially in negative screening experiments to detect dissociated protein complexes. This method utilizes yeast strains expressing toxic hybrid proteins (DB-X/AD-Y or DBPX), where under certain conditions, the dissociation of interacting complexes allows some colonies to grow, and protein interactions can be identified by colony growth status.
