Description

Elastase, Human leukocytes is a serine protease present in the nitrogen-loving granules of neutrophils. Elastase, Human leukocytes potential substrates include almost all components of the extracellular matrix, as well as a variety of proteins such as coagulation factors, complement, immunoglobulins and cytokines. It has a strong proteolytic function and participates in the pathogenesis of inflammatory tissue damage[1][2].

In Vitro

Elastase, Human leukocytes hydrolysis IL-36Ra experiment[2]

(1) Cells were grown in serum-free medium or Opti-MEM medium to prevent any inhibition of active proteases. After isolation, neutrophils were immediately stimulated with 100 ng/mL PMA at 37°C for 1 hour, then centrifuged to collect the cells and supernatant.
(2) 1-2 μg of SUMO-tagged IL-36Ra was incubated with the supernatant from primary cells at 37°C for 1 hour, with or without protease inhibitors (IAA 50 μM, EDTA 500 μM, PMSF 1 mM, 1x Roche protease inhibitor mixture, α1-antitrypsin 1 mg/ml).
(3) Incubate with Elastase, Human leukocytes at 37°C for 5 hours.
(4) Analyze using SDS-PAGE or Western Blotting with a monoclonal anti-IL-36Ra antibody.
