EP-3945 is an agonist of Mas-related G protein-coupled receptor (MRGPR) with greater potency than the MRGPRD small molecule agonist β-Alanine (HY-N0230). MRGPR plays an important role in inflammatory itch and pain sensation. At the same time, MRGPR binds to Gq (MRGPRX2, MRGPRX4, MRGPRX1 are coupled to Gq; MRGPRX2, MRGPRD are coupled to Gi), and the EC50 of EP-3945 on Gq is 211.6 nM.
