Apoptosis is a form of programmed cell death. Based on the cellular changes during programmed cell death, apoptosis is often classified into early, middle, and later stages. The later stages of apoptosis are characterized by changes in nuclear morphology, chromatin condensation, nuclear envelope degradation, and DNA fragmentation.

Annexin V is a Ca2+ - dependent phospholipid-binding protein that has a high affinity for PS. In normal live cells, Phosphatidylserine (PS) is located on the inner side of the cell membrane. Upon initiation of apoptosis, PS translocates from the inner to the outer leaflet of the membrane and binds with Annexin V. Therefore, Annexin V serves as one of the sensitive indicator of early-stage apoptosis. Once labeled with a fluorescent probe (FITC), Annexin V can be used to detect the early-stage apoptosis using fluorescence microscopy or flow cytometry. 7-Aminoactinomycin D (7-AAD) is a cell-membrane impermeable dye that can stain late-stage apoptosis cells and necrosis cells with red fluorescence.

MCE Annexin V-FITC/7-AAD Apoptosis Detection Kit can be utilized to detect the occurrence of early apoptosis. After co-staining with Annexin V (labeled with FITC) and 7-AAD, normal cells are essentially non-fluorescent (Annexin V-/7-AAD-), early-stage apoptosis cells show green fluorescence (Annexin V+/7-AAD-), late-stage apoptosis cells and necrosis cells show green and red fluorescence (Annexin V+/7-AAD +).

Application

1. Apoptosis is a rapid process. For best results, it is recommended to analyze samples within 1 h after staining.

2. The digestion solution should not contain EDTA, as EDTA can interfere with the binding of Annexin V to PS.

3. After trypsin digestion of adherent cells, it is recommended to let them recover in optimal culture conditions and medium for 30 min before staining to avoid false positives.

4. The optimal concentration of the dye is determined by the specific experimental requirements.

5. Fluorescent dyes are subject to quenching, so please avoid light exposure to slow down the fluorescence quenching.

6. If the sample is derived from blood, it's essential to remove the platelets. This is because platelets contain PS, which can bind with Annexin V and interfere with the experimental results.

7. This product is for R&D use only, not for drug, household, or other uses.

8. For your safety and health, please wear a lab coat and disposable gloves to operate.

  • Do most proteins show cross-species activity?

    Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
