MCE Lentivirus Solution Buffer adopts a HEPES and sodium bicarbonate buffering system, containing L-glutamine, trace elements, cryoprotectants, and other components. It is suitable for resuspending lentiviral particles after ultracentrifugation, PEG precipitation, or ultrafiltration concentration.

Lentiviral particles resuspended with this buffer exhibit low cytotoxicity, long-term storage stability, and resistance to multiple freeze-thaw cycles.

Protocol
Resuspension of Lentiviral Particles After Ultracentrifugation or PEG Precipitation

1. Based on the desired concentration factor, add an appropriate amount of lentivirus solution buffer and gently pipette up and down to resuspend the pellet.

Note: For example, if 100 mL of lentiviral supernatant is expected to be concentrated 100-fold, add 1 mL of lentivirus solution buffer.

2. Incubate at room temperature for 4 - 6 h to allow the lentiviral particles to fully release into the solution buffer.

3. Centrifuge at 4°C, 11,200 g (12,000 rpm) for 3 min and collect the supernatant. Aliquot appropriately and store at -80°C.

Note: The above centrifugation is based on a small centrifuge (effective radius ~ 7 cm) and ≤ 2 mL centrifuge tubes. For larger volumes, it is recommended to centrifuge in aliquots.

 

Buffer Exchange After Ultrafiltration Concentration

After ultrafiltration of the lentiviral supernatant, add an appropriate amount of lentivirus solution buffer according to experimental requirements, centrifuge to the desired volume, aliquot appropriately, and store at -80°C.

  • Do most proteins show cross-species activity?

    Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
