MCE Basement Membrane Matrix for Organoid Culture is primarily composed of natural basement membrane matrix extracted from mouse tumors. This product is mainly used for organoid culture.

Protocol
Thin Gel Method

For cell migration and invasion, primary cell expansion: Dilute with pre-cooled medium according to experimental needs after thawing. It is recommended that the concentration be no less than 1 mg/mL. Add 50 μL/cm2 of matrix to the surface of the cell culture plate and spread it evenly, taking care to avoid air bubbles. Place the plate at 37 ℃ and incubate for 0.5 h, aspirate the supernatant to use.

Thin Coating Method.

For primary cell expansion: Dilute with pre-cooled medium according to experimental needs after thawing. It is recommended that the concentration be no less than 0.1 mg/mL. Aspirate the appropriate amount of dilution to completely cover the surface of the cell culture plate, the recommended amount of coating is 0.01-0.02 mg/cm2. Place the plate at 37 ℃ and incubate for 1h, aspirate the supernatant to use.

Gel embedding Method.

For organoid culture and tumor spheroid invasion: Thaw the product in advance and set aside. Prepare the desired cells and resuspend them with matrix, it is recommended that the proportion of matrix is >70%. Add 15-20 μL/cm2 to the surface of the cell culture plate, taking care to avoid air bubbles. Place the plate at 37 ℃ for 0.5 h to form a gel that wraps the cells, and add the appropriate medium to the plate.

 

Note: When thawing the product for the first time, immerse the vial in crushed ice and place it in a 4°C to melt. After melting, mix thoroughly before aliquoting, and then store at -20°C.

  • Do most proteins show cross-species activity?

    Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
