Basement Membrane Matrix GFR

Cat. No.: HY-K6004
Manual SDS

MCE Basement Membrane Matrix GFR is primarily composed of a natural basement membrane matrix derived from mouse tumors and contains less various cytokines than the standard type. This product is mainly used for organoid culture. It is also for research applications with high requirements for basement membrane preparation and has no effect on subsequent experiments requiring color detection.

MCE Biological Validation

Organoid Culture
Gastric cancer organoid culture test: the organoids exhibited consistent characteristics across three consecutive passages, presenting a typical vesicular structure.
Mouse small intestine organoid culture: The low-growth-factor Basement Membrane Matrix ( HY-K6004) allows for stable cultivation of organoids.

    Description
    & Advantages

MCE Basement Membrane Matrix GFR is a natural basement membrane matrix extracted from mouse tumors and is composed mainly of various growth factors and extracellular matrix components. The main extracellular matrix components are: Laminin, Col-IV, Entactin, Heparan sulphate proteoglycans, etc. The main growth factor components are: insulin-like growth factor (IGF-1), transforming growth factor beta (TGF-β), vascular endothelial growth factor (VEGF), epidermal growth factor (EGF), fibroblast growth factor (bFGF), etc. The percentage of each of these growth factors is lower than that of standard basement membrane matrix.

MCE Basement Membrane Matrix GFR (Phenol Red) reduces the background signal induced by growth factors and is suitable for organoid culture. It is also for research applications with high requirements for basement membrane preparation and has no effect on subsequent experiments requiring color detection.

    Storage

    -20°C, 2 years.

    Avoid repeated freezing and thawing.

    Protocol

    a. Use a pre-cooled pipette to blow the matrix gel evenly after thawing.

    b. Use pre-cooled serum-free medium to dilute according to experimental requirements. Refer to the Experimental Conditions table for final concentrations.

    c. Place the culture plate on ice and inject the substrate gel dilution slowly against the wall to avoid air bubbles. Refer to the Experimental Conditions table for the volume injected.

    d. Transfer the culture plate to 37℃ incubator gently. After gelling, aspirate off excess supernatant. Refer to the Experimental Conditions table for gel time.

Note: When thawing the product for the first time, immerse the vial in crushed ice and place it in a 4°C to melt. After melting, mix thoroughly before aliquoting, and then store at -20°C.

    Help & FAQs
    • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
