Alkaline Phosphatase/ALPL Protein is a preproprotein that is proteolytically cleaved to yield a signal peptide and a proproptein that is subsequently processed to generate the active mature peptide. ALPL is also a membrane-bound glycosylated enzyme that catalyzes the hydrolysis of phosphate esters at alkaline pH. Mice that lack ALPL show symptoms of osteomalacia, softening of the bones. Moreover, ALPL stimulates mineralization mainly through modulation of the balance between inorganic phosphate (Pi) and inorganic pyrophosphate (PPi) and also has a role in cardiovascular remodelling. Background

Alkaline phosphatase (ALPL) is a preproprotein that is proteolytically cleaved to yield a signal peptide and a proproptein that is subsequently processed to generate the active mature peptide. ALPL is also a membrane-bound glycosylated enzyme that catalyzes the hydrolysis of phosphate esters at alkaline pH. The mature peptide maintains the ratio of inorganic phosphate to inorganic pyrophosphate required for bone mineralization. Mice that lack ALPL show symptoms of osteomalacia, softening of the bones. In humans, mutations in this gene are associated with hypophosphatasia, an inherited metabolic bone disease in which deficiency of this enzyme inhibits bone mineralization leading to skeletal defects. ALPL is highly expressed in the cells of mineralized tissue and plays a critical function in the formation of hard tissue. And ALPL increases inorganic phosphate local rates and facilitates mineralization as well as reduces the extracellular pyrophosphate concentration, an inhibitor of mineral formation. Moreover, ALPL stimulates mineralization mainly through modulation of the balance between inorganic phosphate (Pi) and inorganic pyrophosphate (PPi) and also has a role in cardiovascular remodelling[1][2][3].
