Background

Ang2, a 496 amino acid-long protein, shares ~60% amino acid homology with Ang1 and lacks one of the nine cysteines found in mature Ang1. It has a secretion signal peptide, an NH2-terminal coiled-coil domain, and a COOH-terminal fibrinogen-like domain. Unlike Ang1, Ang2 acts in an autocrine manner and its expression is highly regulated. Similar to Ang1, Ang2 binds to the Tie2 receptor with the same binding affinity, inducing its antagonistic role, but does not bind to Tie1. Ang2 expression is triggered by inflammatory mediators, such as thrombin [4], and conditions, such as hypoxia and cancer[1].
ANGPT2 has been described as a context-dependent antagonist interfering with angiopoietin-1-induced Tie2 phosphorylation to destroy vascular stability and promote angiogenesis, might confer resistance to antiangiogenic therapy. ANGPT2 has been found to be highly expressed in diverse tumor cells and plays an important role in tumor angiogenesis and inflammation[2].
