Background

The Leptin R/LEPR Protein, a member of the gp130 family of cytokine receptors, activates gene transcription through the cytosolic STAT proteins. Serving as a receptor for leptin—an adipocyte-specific hormone crucial for body weight regulation—this protein plays a pivotal role in the regulation of fat metabolism and contributes to a novel hematopoietic pathway essential for normal lymphopoiesis. Mutations in this gene have been associated with obesity and pituitary dysfunction, underscoring its significance in metabolic and endocrine functions. Notably, alternative splicing produces transcript variants encoding different isoforms, and it is noteworthy that this gene shares the same promoter and the first two exons with the LEPROT gene, albeit encoding distinct proteins. The gene displays ubiquitous expression, with elevated levels observed in the liver (RPKM 22.2), endometrium (RPKM 12.0), and 23 other tissues, indicating its diverse roles across various physiological contexts in multiple organs.

Biological Activity

Measured by its binding ability in a functional ELISA. Immobilized human Leptin at 1.25 μg/mL (100 μL/well) can bind Biotinylated human Leptin receptor. The ED50 for this effect is 0.1843 μg/mL.

Reconstitution

Stored at -20°C for 2 years. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
