Background

Neuropilin-1 Protein is a cell-surface receptor that plays diverse roles in the development of the cardiovascular system, angiogenesis, neuronal circuit formation, and organogenesis. It acts as a mediator for the chemorepulsant activity of semaphorins and recognizes a C-end rule (CendR) motif R/KXXR/K on its ligands, leading to cellular internalization and vascular leakage. It binds to various ligands including semaphorin 3A, PLGF-2 isoform of PGF, VEGF165 isoform of VEGFA, and VEGFB. When coexpressed with KDR, it enhances the binding of VEGF165 to KDR and promotes chemotaxis. Neuropilin-1 regulates VEGF-induced angiogenesis and plays a crucial role in motor neuron axon guidance and cell body migration, particularly during the caudal migration of facial motor neurons in embryonic development. It is also involved in the regulation of mitochondrial iron transport through its interaction with ABCB8/MITOSUR. Neuropilin-1 forms homodimers and heterodimers with NRP2, interacts with PLXNB1, FER, VEGFA, and ABCB8/MITOSUR in mitochondria.

Biological Activity

Measured by its binding ability in a functional ELISA. Immobilized Rat Neuropilin-1 at 2 µg/mL can bind Human VEGF165. The ED50 for this effect is 0.8699 nM.
