Background

Wee1 Protein functions as a critical negative regulator of the G2 to M transition, preventing entry into mitosis by safeguarding the nucleus from the activation of cytoplasmically bound cyclin B1-complexed CDK1. Through the phosphorylation of CDK1 on 'Tyr-15,' Wee1 specifically targets and inactivates the cyclin B1-CDK1 complex, with its activity peaking during the G2 phase and reaching a minimum as cells progress into M phase. Notably, Wee1's phosphorylation of cyclin B1-CDK1 occurs exclusively on 'Tyr-15,' while monomeric CDK1 remains unphosphorylated.
