Description

Bicine is a buffer used in physiological research. Bicine is a derivative of the simple amino acid glycine that increases the sensitivity of guanosine enzyme histochemical detection[1][2].

In Vitro

Preparation of sections for histochemistry of guanine oxidase in human tissues[2]:
1. Tissue samples were snap frozen in liquid nitrogen and 10 µM thick cryostat sections were prepared.
2. After fixation with 2.5% glutaraldehyde in 0.05 M sodium cacodylate buffer (pH 7.4) at room temperature for 1 hour, sections were rinsed in Bicine buffer (0.1 M, pH 7.8) and incubated with xanthine oxidase at 37°C for 15 minutes, rinsed again in Bicine buffer and incubated at 37°C for 1-2 hours.
3. After rinsing in distilled water, sections were allowed to air dry, dehydrated with graded ethanol, transferred to xylene and mounted in synthetic resin.
