Description

Borocaptate sodium is a boron-containing compound that can be used as a therapeutic agent in boron neutron capture therapy (BNCT), exhibiting anticancer activity[1][2].

In Vitro

Borocaptate sodium (60 μg 10B/mL, 24 hours) combined with BSO (HY-106376) enhances boron uptake and inhibits cell viability in mouse squamous carcinoma cells, rat glioma cells, and hamster lung cells, with boron uptake increasing as cell viability decreases[3].

Cell Viability Assay[3]

Cell Line: Mouse squamous cell carcinoma SCCVII, rat glioma cell line C6, Chinese hamster V79 cells.
Concentration: 60 μg 10B/mL (Each milliliter of Borocaptate sodium solution contains 60 µg of boron-10 isotope (10B))
Incubation Time: 24 h
Result: Increased boron uptake in tumor cells by 19.2%-34.4% and enhanced the inhibitory effect on cell viability (combined with BSO (HY-106376), a glutathione inhibitor).
In Vivo

Borocaptate sodium (55 mg/kg, i.v., single dose) selectively targets intracranial tumors in dogs and inhibits their growth[2].

Animal Model: Spontaneous intracranial tumor-bearing dogs (primarily gliomas, mixed breed dogs, typically between 5-10 years old, weight 20-30 kg)[2]
Dosage: 55 mg/kg
Administration: Intravenous injection (i.v.), single dose
Result: Resulted in boron concentrations of 35.9 µg/g in the tumor at 2 hours, 22.5 µg/g at 6 hours, and 7.0 µg/g at 12 hours. The boron concentration in the tumor was higher than that in normal brain tissue, with a favorable tumor-to-brain ratio.
