Cationic Bovine Serum Albumin  (Synonyms: CBSA)

Cat. No.: HY-NP001 Purity: ≥95.0%
Handling Instructions Technical Support

Cationic Bovine Serum Albumin is a bovine serum albumin (BSA) modified with excess ethylenediamine, which has enhanced immunogenicity compared to native BSA. Positively charged primary amines block all negatively charged carboxyl groups, making Cationic Bovine Serum Albumin a cationic protein. Cationic Bovine Serum Albumin has remarkable neuroprotective effects on ischemic stroke when in combination with Tanshinone IIA (HY-N0135) .

Description

Cationic Bovine Serum Albumin is a bovine serum albumin (BSA) modified with excess ethylenediamine, which has enhanced immunogenicity compared to native BSA. Positively charged primary amines block all negatively charged carboxyl groups, making Cationic Bovine Serum Albumin a cationic protein. Cationic Bovine Serum Albumin has remarkable neuroprotective effects on ischemic stroke when in combination with Tanshinone IIA (HY-N0135) [1].

In Vivo

Cationic Bovine Serum Albumin can be used in animal modeling to create urinary system disease models[1][2].

Induction of Membranous Nephropathy[1][2]
Background
Cationic Bovine Serum Albumin induced diffuse glomerular basement membrane (GBM) thickening and a predominant T-helper (Th) 2 type immune response, followed by glomerular capillary wall (GCW) damage and severe proteinuria, finally progressing to renal function impairment[1].
Specific Modeling Methods
Mice: SPF ICR, BALB/c and C57BL/6• female• 8 weeks of age• weighing 20 g[1]
Administration: 1-9 mg/kg • i.v.• a single dose for 4 weeks[1]
Note
(1) 200g of crystallized bovine serum albumin was dissolved in 1.2 mL of distilled water. Anhydrous ethylenediamine (EDA) was prepared by mixing 2.68 mL of EDA and 20 mL of distilled water. The pH was adjusted to 4.75 with 6N HCl and the solution cooled to 25°C. This EDA solution was added slowly to the BSA solution followed by 150 mg of 1-ethyl-3-[(3-dimethylaminopropyI)-carbodimide hydrochlor-ide] (EDC) with continuous stirring for 4h. The product was purified using the pH-dependent binding technique[1].
(2) ICR mice received the nephritogenic antigen intravenously at 1, 3, 5, 7 and 9 mg/kg, three times per week every other day, two weeks after immunization by the same route with 0.2 mg of Cationic Bovine Serum Albumin emulsified in anequal volume of complete Freund's adjuvant (HY-153808)[1].
Modeling Indicators
Metabolic change: Cationic Bovine Serum Albumin administration developed hyperlipidaemia, hypoalbuminaemia and severe proteinuria[1].
Morphological changes: Cationic Bovine Serum Albumin administration exhibited diffuse thickening of the GBM and no significant mesangial proliferation[1].
Molecular changes: Cationic Bovine Serum Albumin administration H mice showed strong granular fluorescent intensity of IgG and C3 along the glomerular capillary wall (GCW). The Th2 type immune response predominated in mice[1].
Pathology change: The basement membrane showed severe irregular thickening of the laminae rarae, interna and externa and subepithelial deposits were layered and more confluent, suggesting glomerular size selectivity defect[1].

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
