Description

CHIKV-IN-5 (Compound 26) is a CHIKV inhibitor (EC90 = 0.45 μM). CHIKV-IN-5 inhibits CHIKV replication at a late stage in the virus life cycle by blocking structural protein translation. CHIKV-IN-5 has great in vitro mouse microsomal stability. CHIKV-IN-5 reduces footpad swelling and decreases virus dissemination to other tissues in mice infected with CHIKV[1].

In Vitro

CHIKV-IN-5 shows good virus titer reduction (VTR = 8.7 log at 10 μM concentration) in normal human dermal fibroblast cells and in vitro mouse microsomal stability (MLMt1/2=74 min)[1].
CHIKV-IN-5 (10 μM, 2 h) blocks CHIKV replication post-entry on NHDF cells[1].
CHIKV-IN-5 (10 μM, pretreats for 1 h) inhibits CHIKV structural protein synthesis but not viral transcription in CHIKV infected NHDF cells [1].

In Vivo

CHIKV-IN-5 (80 mg/kg, i.p., twice a day, 3 days) reduces footpad swelling and significantly decrease virus dissemination to other tissues in CHIKV-infected mice [1].
CHIKV-IN-5 (0.5-20 mg/kg, i.p., i.v., p.o., s.c.) shows favorable metabolic stability in male C57BL/6 mice [1].
