Description

CMP-sialic acid synthetase (NmCSS) is an essential enzyme involved in the biosynthesis of carbohydrates and glycoconjugates containing sialic acids. CMP-sialic acid synthetase (NmCSS) activates free Sia, converting it to CMP-Sia, which is the only donor substrate for all sialyltransferases[1].

In Vitro

Product Information This enzyme is used to activate sialic acid to form CMP-sialic acid. It serves as a donor for sialic acid transfer by glycosyltransferase and can be used as a tool enzyme for sialic acid sugar transfer. This product is recombinantly expressed in Escherichia coli, with a C-terminal His Tag, a molecular weight of 26 kDa, and a storage buffer component of 20 mM Tris-HCl, pH 8.0, 5% glycerol.

Instructions
1. Add substrate sialic acid and CTP according to the reaction system. Generally, the concentration is controlled below 100 mM. Buffer salts (such as Tris-HCl) can be appropriately added to the reaction system;
2. Add 5 mM MgCl2 to the reaction system, mix well, adjust the pH to 7.5-8.5, and add an appropriate amount of enzyme (according to the reaction requirements).
3. Incubate the sample at 37℃ for 30-60 min. The recommended reaction buffer is Tris-HCl (pH 8.0), and most common biological buffers are also applicable, such as Tris or PBS.

Notes
When the system is scaled up, the incubation time or enzyme amount needs to be adjusted according to the actual situation. It can be mixed by low-speed oscillation. Adding Inorganic pyrophosphatase, Saccharomyces cerevisiae (HY-P2992) can increase the conversion rate.
