Description

Endo-β-N-acetylglucosaminidase (Endo S2) is a key enzyme involved in the processing of free oligosaccharides in the cytosol. Endo-β-N-acetylglucosaminidase catalyzes hydrolysis of N-linked oligosaccharides[1].

Endo S2 is a highly specific endoglycosidase that can cleave high-mannose, hybrid, and dual-antenna complex N-glycans with or without core fucosylation from IgG. It is also possible to transfer the oxazoline form of N-glycans to antibody N-glycosylation sites after hydrolysis with Endo S2, with or without core fucose.
This product is expressed by recombinant E. coli, C-terminal His Tag, molecular weight 92 kDa, storage buffer is 20 mM Tris-HCl, pH 7.5, 200mM NaCl, 10% glycerol, which can be used for antibody glycosylation modification and analyze.

Instructions
1. Dissolve 1-20 μg of glycoprotein in deionized water, add 1 μL of 500 mM Tris-HCl (pH7.5), and adjust the volume to 10 μL with deionized water;
2. Add 1-2 μL of Endo S2, pipe gently to mix;
3.37 ℃ incubation 60-120 minutes;
4. Used for SDS-PAGE analysis or mass spectrometry analysis.
Note: Most common biological buffers are suitable, such as Tris or PBS. Buffers outside this pH range (such as acetate buffer) may be suitable, but the incubation time or enzyme amount needs to be optimized according to the actual situation.
