Description

FerroOrange is a fluorescent probe for the detection of unstable divalent iron ions, localized in the endoplasmic reticulum, which emits irreversible orange fluorescence upon encountering divalent iron ions. Excitation/emission wavelength: 542/572 nm.

In Vitro

Solution preparation
Add 35 µL DMSO to the tube containing 24 µg FerroOrange and mix well with a pipette to prepare a 1 mM FerroOrange solution.
Dilute the 1 mM FerroOrange solution with HBSS solution to prepare a 1 µM FerroOrange working solution.
Note:
1. The 1 mM FerroOrange stock solution can be stored for 1 month at -20°C. The FerroOrange working solution is unstable. It needs to be prepared and used as soon as possible.
2. If necessary, serum-free culture medium can be used instead of HBSS. Serum will increase the background.
Procedure
1. Inoculate cells in a fluorescent culture dish and culture overnight in a 37°C, 5% CO2 incubator.
2. Discard the supernatant and wash the cells 3 times with HBSS or serum-free medium.
3. Replace the medium containing drugs and culture in a 37℃, 5% CO2 incubator.
Note: The culture time can be adjusted according to actual conditions.
4. Remove the medium and wash 3 times with HBSS or serum-free medium.
5. Add FerroOrange working solution with a concentration of 1 µM and culture in a 37℃, 5% CO2 incubator for 30 minutes.
Note: No need to wash, observe directly after culture.
6. Observe the cells under a fluorescence microscope.
