Indusatumab vedotin (MLN-0264; AKT-264) is an antibody-drug conjugate (ADC) consisting of the human anti-guanylate cyclase C (GCC) antibody linked to a microtubule-disruptor Monomethyl auristatin (HY-15162). Indusatumab vedotin has anti-proliferative effects on pancreas cancer cell lines. Indusatumab vedotin arrests cell cycle at G2/M phase, and induces apoptosis[1].

Guanylate cyclase (GCC)[1]

In Vitro

Indusatumab vedotin (0.4-25 μg/mL; 72 h) causes antiproliferative effects in pancreatic cancer cell lines[1].
Indusatumab vedotin (12.5 μg/mL; 24 h) arrests cell cycle at G2/M in MIA PaCa-2 and PANC-1 cells, and causes the P-Hitone H3 production[1].

Western Blot Analysis[1]

Cell Line: MIA PaCa-2 and PANC-1 cells

Incubation Time: 24 hours
Result: Increased the protein level of P-Hitone H3, indicating tubulin polymerization.

Cell Cycle Analysis[1]

Cell Line: MIA PaCa-2 and PANC-1 cells
Concentration: 12.5 μg/mL
Incubation Time: 24 hours
Result: Decreased cell number in G1 phase and caused an induction of a G2/M arrest.

Western Blot Analysis[1]

Cell Line: GCC Knock cells and Pancreatic cancer cell lines: L3.6, MIA PaCa-2, Panc 03.27, Panc 05.04 and Panc 02.03
Concentration: 0.4 μg/mL, 0.8 μg/mL, 1.6 μg/mL, 3.1 μg/mL, 6.3 μg/mL, 12.5 μg/mL, and 25 μg/mL
Incubation Time: 72 hours
Result: Inhibited the cell viability of pancreatic cancer cells, but not GCC Knock cells.
In Vivo

Indusatumab vedotin (10 mg/kg; i.v.; once weekly for 17 days at least) dramatically decreased the growth rate of tumor[1].
Indusatumab vedotin shows strongly correlated biological activity with exposure in mice bearing one of six different xenograft models[2].
