& Advantages

Hypoxanthine and thymidine supply purines and pyrimidines for DNA synthesis by hybridoma cells via the salvage pathway that utilizes HGPRT contributed by the fused spleen cell. Unfused HGPRT-deficient myeloma cells cannot replicate DNA to proliferate, and unfused spleen cells are mortal, allowing for specific selection of HGPRT-myeloma hybridoma cells. The aminopterin inhibits the de novo nucleoside biosynthesis pathway, after the hybridomas have undergone selection and isolation, aminopterin should be removed.

MCE HT Supplement (50×) is a liquid mixture of hypoxanthine (5 mM) and thymidine (0.8 mM), and can be added to cell culture medium as an additive for the DNA salvage synthesis pathway. It is suitable for post-selection rescue to overcome the effects of residual intracellular aminopterin on the classical DNA synthesis pathway. Once the de novo biosynthesis pathway for nucleosides has been reestablished, HT supplement can be discontinued.

Application

1. This product is a 50× stock solution. Please dilute it as needed before use.

2. This product appears as a colorless and transparent liquid, with an endotoxin content of ≤ 0.25 EU/mL.

3. Precipitation may occur after thawing, dissolve it completely in a 37°C water bath.

4. It is recommended to appropriately aliquot the solution after thawing to avoid repeated freeze-thaw cycles.

5. Pay attention to aseptic operation to avoid contamination.

6. This product is for R&D use only, not for drug, household, or other uses.

7. For your safety and health, please wear a lab coat and disposable gloves to operate.

Protocol

1. Thaw MCE HT Supplement (50×) at room temperature in advance.

2. Take the appropriate amount of HT Supplement (50×) in a sterile environment and add it to the cell culture medium at a 50-fold dilution according to the experimental requirements. For example: add 1 mL of supplement to 49 mL of medium.
