Background

The AGRP protein plays a crucial role in maintaining weight homeostasis. It is involved in the regulation of feeding behavior through the central melanocortin system. AGRP acts as an antagonist of alpha melanocyte-stimulating hormone by inhibiting cAMP production mediated by the stimulation of melanocortin receptors in the hypothalamus and adrenal gland. It exhibits minimal activity with MC5R but functions as an inverse agonist for MC3R and MC4R, effectively suppressing their constitutive activity. AGRP also promotes the endocytosis of MC3R and MC4R in an arrestin-dependent manner. Furthermore, it interacts with melanocortin receptors MC3R, MC4R, and MC5R.
