Description

Annexin A2/ANXA2 Protein, Human is a 38kDa multifunctional protein which belongs to the annexin family. Annexin A2 is important for the regulation in the cellular growth and in signal transduction pathways. Annexin A2 also can be used as an autocrine factor that can heighten osteoclast formation and bone resorption[1].

Background

The Annexins comprise a family of proteins that are involved in many aspects of cellular membrane dynamics and the regulation of membrane-associated proteins.
Annexin A2 is an important member of annexin family of proteins expressed on surface of endothelial cells (ECs), macrophages, mononuclear cells and various types of cancer cells. Annexin A2 exhibits high affinity binding for calcium (Ca2+) and phospholipids. ANX A2 plays an important role in many biological processes such as endocytosis, exocytosis, autophagy, cell-cell communications and biochemical activation of plasminogen.

Biological Activity

Measured in a cell proliferation assay using A549 cells. The ED50 this effect is ≤1.051 ng/mL, corresponding to a specific activity is ≥9.515×105 units/mg.

  • Measured in a cell proliferation assay using A549 cells. The The ED50 for this effect is 0.7184 ng/mL, corresponding to a specific activity is 1.39×106 units/mg.

Approximately 38 kDa

Purity
  • Greater than 95% as determined by reducing SDS-PAGE.
Appearance

Lyophilized powder.

Formulation

Lyophilized after extensive dialysis against 20 mM Histidine-HCl, 6% Sucrose, 4% Mannitol, 0.05% Tween 80, pH 5.5. or PBS, pH 7.4 or PBS, pH 7.4, 6% saccharose, 4% mannitol and 0.05% Tween80.

Endotoxin Level

<1 EU/μg, determined by LAL method.

Reconstitution

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Storage & Stability

Stored at -20°C for 2 years. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.

Shipping

Room temperature in continental US; may vary elsewhere.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Dilution Calculator

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
