APAF1 Protein, Human (P. pastoris, His)

Cat. No.: HY-P700502
COA Handling Instructions

APAF1 Protein, a pivotal apoptosome component, orchestrates cytochrome c-dependent activation of pro-caspase-9, initiating caspase-3 activation and apoptosis. Forming an apoptosome ring with cytochrome c and dATP, APAF1 interacts with pro-caspase-9, releasing mature caspase-9. Pro-caspase-3 joins the complex, activating caspase-3. Notably, APAF1 isoform 6 is less effective in inducing apoptosis. APAF1 interacts with APIP, NAIP/BIRC1, and CIAO2A, contributing to its multifaceted role in apoptotic regulation. APAF1 Protein, Human (P. pastoris, His) is the recombinant human-derived APAF1 protein, expressed by P. pastoris, with N-6*His labeled tag. The total length of APAF1 Protein, Human (P. pastoris, His) is 312 a.a., with molecular weight of 37.4 kDa.

Background

APAF1 Protein, as an essential component of the apoptosome, orchestrates the cytochrome c-dependent autocatalytic activation of pro-caspase-9 (Apaf-3), initiating the caspase-3 activation cascade and ultimately leading to apoptosis. The oligomeric form of APAF1 forms a heptameric ring, known as the apoptosome, in the presence of cytochrome c and dATP. Within this complex, APAF1 and pro-caspase-9 interact via their NH2-terminal CARD domains, leading to the release of mature caspase-9. Concurrently, pro-caspase-3 is recruited into the APAF1-pro-caspase-9 complex through interaction with pro-caspase-9, facilitating the activation of caspase-3. Notably, isoform 6 of APAF1 exhibits reduced effectiveness in inducing apoptosis. Additionally, APAF1 interacts with APIP, NAIP/BIRC1, and CIAO2A, contributing to its multifaceted role in the regulation of apoptotic pathways.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Dilution Calculator

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
