Background

CCDC47 Protein is a crucial component of the multi-pass translocon (MPT) complex, which facilitates the insertion of multi-pass membrane proteins into the lipid bilayer of membranes. Following initial membrane insertion by the SEC61 complex, the MPT complex takes over and blocks the lateral gate of the SEC61 complex, promoting the insertion of subsequent transmembrane regions. Within the MPT complex, the PAT subcomplex, consisting of CCDC47, plays a role in sequestering highly polar regions in the transmembrane domains away from the non-polar membrane environment until they can be properly buried in the fully assembled protein. Additionally, CCDC47 occludes the lateral gate of the SEC61 complex within the PAT subcomplex. CCDC47 is also involved in regulating calcium ion homeostasis in the endoplasmic reticulum (ER) and is essential for proper protein degradation via the ER-associated degradation (ERAD) pathway. Its presence is necessary for maintaining ER organization during embryogenesis. CCDC47 forms the PAT complex along with WDR83OS/Asterix and is part of the larger MPT complex, which includes the GEL complex (RAB5IF/OPTI and TMCO1), the BOS complex (NCLN/Nicalin, NOMO, and TMEM147), and the PAT complex. The MPT complex interacts with the SEC61 complex and also interacts with VCP, HSPA5, DERL1, DERL2, and SELENOS.

Reconstitution

Stored at -20°C for 2 years. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
