Granzyme B/GZMB Protein, Mouse (HEK293, His, solution)

Cat. No.: HY-P70253
SDS COA Handling Instructions Technical Support

Granzyme B/GZMB Protein, found in cytotoxic T-cells and NK-cells, activates caspase-independent pyroptosis and cleaves gasdermin-E (GSDME), inducing target cell death. It also participates in caspase activation cascade, promoting apoptosis and plasma membrane repair during bacterial infection. Granzyme B/GZMB Protein, Mouse (HEK293, His, solution) is the recombinant mouse-derived Granzyme B/GZMB protein, expressed by HEK293 , with C-6*His labeled tag.

Description

Granzyme B/GZMB Protein, found in cytotoxic T-cells and NK-cells, activates caspase-independent pyroptosis and cleaves gasdermin-E (GSDME), inducing target cell death. It also participates in caspase activation cascade, promoting apoptosis and plasma membrane repair during bacterial infection. Granzyme B/GZMB Protein, Mouse (HEK293, His, solution) is the recombinant mouse-derived Granzyme B/GZMB protein, expressed by HEK293 , with C-6*His labeled tag.

Background

Granzyme B/GZMB Protein, an abundant protease found in the cytosolic granules of cytotoxic T-cells and NK-cells, plays a crucial role in various cellular processes. When delivered into the target cell through the immunological synapse, Granzyme B/GZMB activates caspase-independent pyroptosis, leading to target cell death. It achieves this by cleaving after Asp and catalyzing the cleavage of gasdermin-E (GSDME), releasing the pore-forming component of GSDME, which triggers pyroptosis. Granzyme B/GZMB is also involved in the activation cascade of caspases, including caspase-3, -9, and -7, which are responsible for apoptosis execution and plasma membrane repair in response to bacterial infection.

Biological Activity

Measured by its ability to cleave a peptide substrate, t-Butyloxycaronyl-Ala-Ala-Asp-ThioBenzyl ester (Boc-AAD-SBzl), in the presence of 5,5’-Dithio-bis (2-nitrobenzoic acid) (DTNB). The specific activity is 2136.752 pmol/min/µg as measured under the described conditions.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
