HA1/Hemagglutinin Protein, H5N8 (P11135, HEK293, His)

Cat. No.: HY-P73970
Handling Instructions

The HA/Hemagglutinin protein binds to cell surface receptors, facilitating virus attachment. It triggers internalization via endocytosis. HA determines host range and virulence, mediating fusion of virus and endosomal membrane. In endosomes, HA2 undergoes conformational change, releasing fusion peptide and forming fusion pore. Multiple HA trimers are needed for pore formation. HA/Hemagglutinin Protein, H5N8 (P11135, HEK293, His) is the recombinant Virus-derived HA/Hemagglutinin protein, expressed by HEK293 , with C-His labeled tag.

Background

The HA/Hemagglutinin protein binds to sialic acid-containing receptors on the cell surface, facilitating the attachment of the virus particle to the cell. This attachment leads to the internalization of about two thirds of the virus particles via clathrin-dependent endocytosis and about one third via a clathrin- and caveolin-independent pathway. HA also plays a crucial role in determining the host range restriction and virulence of the virus. As a class I viral fusion protein, it is responsible for penetrating the cell cytoplasm by mediating the fusion of the membrane of the endocytosed virus particle with the endosomal membrane. In the low pH environment of endosomes, HA2 undergoes an irreversible conformational change, releasing the fusion hydrophobic peptide and forming a competent fusion pore. Multiple trimers of HA are required for the formation of this pore.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
