Cat. No.: HY-P702130
Handling Instructions

HpGT Protein, Helicobacter pylori is a glutathione-degrading enzyme that is a virulence factor in infection. Glutathione hydrolase proenzyme belongs to the gamma-glutamyltransferase family. pylorigamma-glutamyltranspeptidase is expressed as a 60-kDa inactive precursor that must undergo autocatalytic processing to generate a 40-kDa/20-kDa heterodimer with full gamma-glutamyl amide bond hydrolase activity. HELPY Protein, Helicobacter pylori (His) is the recombinant HpGT protein, expressed by E. coli , with N-6*His labeled tag. The total length of HELPY Protein, Helicobacter pylori (His) is 543 a.a., .

Background

pylorigamma-glutamyltranspeptidase (HpGT) is expressed as a 60-kDa inactive precursor that must undergo autocatalytic processing to generate a 40-kDa/20-kDa heterodimer with full gamma-glutamyl amide bond hydrolase activity. The resultant N terminus, Thr-380, has been shown to be the catalytic nucleophile in both the autoprocessing and enzymatic reactions. HpGT is a member of the N-terminal nucleophile hydrolase superfamily. HpGT has also been shown to up-regulate COX-2 and epidermal growth factor-related peptides in human gastric mucosal cells and to induce apoptosis in human gastric epithelial cells[1].
Helicobacter pylori γ-glutamyltranspeptidase (HpGT)  catalyzes the first step in glutathione degradation, removing glutamate from the tripeptide and its conjugates via a γ-glutamylenzyme intermediate. The γ-glutamyl group can be transferred to water (hydrolysis) or an amino acid or short peptide (transpeptidation)[2].

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
