Background

Heparin-binding protein (HPRG) is a versatile plasma glycoprotein known for its ability to bind various ligands, including heme, heparin, heparan sulfate, thrombospondin, plasminogen, and divalent metal ions. Functioning as an adapter protein, HPRG is implicated in the regulation of diverse physiological processes such as immune complex and pathogen clearance, cell adhesion, angiogenesis, coagulation, and fibrinolysis. Its involvement extends to mediating the clearance of necrotic cells through an enhanced phagocytosis pathway, dependent on heparan sulfate. This process is subject to modulation by specific HPRG ligands, including heparin and zinc ions. HPRG exhibits affinity for IgG subclasses of immunoglobulins with kappa and lambda light chains, playing a regulatory role in their clearance and inhibiting the formation of insoluble immune complexes. Moreover, HPRG tethers plasminogen to cell surfaces and interacts with T-cells, influencing their morphology. Notably, it modulates angiogenesis by counteracting the antiangiogenic effect of thrombospondins, specifically THBS1 and THBS2, through interactions with CD36. Additionally, HPRG engages in interactions with various proteins, including HPSE, TMP1, F12, PLG, and TPM1, contributing to its involvement in diverse cellular and molecular processes.
