Background

IL-2 Protein, produced by activated CD4-positive helper T-cells and to a lesser extent activated CD8-positive T-cells and natural killer (NK) cells, plays vital roles in the immune response and tolerance. It binds to a receptor complex composed of either the high-affinity trimeric IL-2R (IL2RA/CD25, IL2RB/CD122, and IL2RG/CD132) or the low-affinity dimeric IL-2R (IL2RB and IL2RG), leading to downstream signaling through phosphorylation of JAK1 and JAK3. This activation triggers various pathways, including STAT, phosphoinositide-3-kinase/PI3K, and mitogen-activated protein kinase/MAPK pathways. IL-2 Protein acts as a T-cell growth factor, enhancing NK-cell cytolytic activity, promoting the proliferation of activated B-cells, and stimulating immunoglobulin production. It also plays a crucial role in regulating the adaptive immune system by controlling the survival and proliferation of regulatory T-cells, which are essential for immune tolerance. Additionally, IL-2 Protein participates in the differentiation and homeostasis of effector T-cell subsets, such as Th1, Th2, Th17, and memory CD8-positive T-cells.

Biological Activity

1.Measured by its binding ability in a functional ELISA.Immobilized recombinant Canine IL2 at 10 μg/mL can bind recombinant human IL2Ra-Fc with a linear range of 0.3125-20 μg/mL.
2.Measured in a cell proliferation assay using CTLL-2 mouse cytotoxic T cells. The ED50 for this effect is 0.3569 ng/mL, corresponding to a specific activity is 2.8×106 units/mg.
