Background

The MAG/Siglec-4a protein is an adhesion molecule that plays a crucial role in mediating interactions between myelinating cells and neurons. It accomplishes this by binding to neuronal sialic acid-containing gangliosides, as well as the glycoproteins RTN4R and RTN4RL2. While it is not necessary for initial myelination, it appears to be involved in maintaining normal axon myelination and protecting motoneurons from apoptosis, even after injury. The protein is also required to prevent degeneration of myelinated axons in adults, likely through its binding to gangliosides on the axon cell membrane. Additionally, it acts as a negative regulator of neurite outgrowth, inhibiting both axon longitudinal growth and axon outgrowth by binding to RTN4R. Its inhibitory effects on neurite extension are primarily mediated by its interaction with neuronal RTN4R, RTN4RL2, and gangliosides. The MAG/Siglec-4a protein can exist as a monomer or homodimer and interacts with isoform 2 of BSG through its first three N-terminal Ig-like domains.

Biological Activity

Measured by its ability to inhibit proliferation of SH-SY5Y cells. The ED50 for this effect is 2.501 μg/mL.
