Background

Neuroligin-1 (NLGN1), a cell surface protein, actively participates in cell-cell interactions through its engagement with neurexin family members. It plays a pivotal role in synapse function and the transmission of synaptic signals, likely by recruiting and clustering various synaptic proteins. While NLGN1 may contribute to the initial formation of synapses, it is not deemed indispensable for this process. In vitro studies indicate its ability to initiate the de novo formation of presynaptic structures, suggesting involvement in the specification of excitatory synapses. Moreover, NLGN1 is crucial for maintaining the quality of wakefulness and normal synchrony of cerebral cortex activity during wakefulness and sleep. This protein's intricate interactions include binding to neurexins, specifically NRXN1, NRXN2, and NRXN3, mediated by heparan sulfate glycan modification on neurexin. Additionally, NLGN1 interacts with NLGN3, AIP1, PDZRN3, DLG4/PSD-95 (via PDZ domain 3), and GOPC, collectively contributing to its role in nervous system development.

Biological Activity

Measured by its binding ability in a functional ELISA. When recombinant human Neuroligin-1 is immobilized at 5 μg/mL, biotinylated recombinant human Neurexin 1 beta binds with an apparent KD is 36.43 nM.

Reconstitution

Stored at -20°C for 2 years. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
