Background

NOV/CCN3 protein is involved in various biological processes including migration, differentiation, survival, and inflammation. It acts by binding to integrins or membrane receptors such as NOTCH1. It is an essential regulator of hematopoietic stem and progenitor cells and inhibits myogenic differentiation through the activation of the Notch-signaling pathway. It also inhibits vascular smooth muscle cell proliferation and supports endothelial cell adhesion, migration, and survival, promoting angiogenesis. In cutaneous wound healing, it acts as an integrin receptor ligand, supporting fibroblast adhesion and chemotaxis. It is involved in bone regeneration and enhances the articular chondrocytic phenotype while repressing endochondral ossification. It impairs pancreatic beta-cell function and plays a role as a negative regulator of endothelial pro-inflammatory activation, reducing monocyte adhesion and inhibiting NF-kappaB signaling pathway. Additionally, it contributes to the control and coordination of inflammatory processes in atherosclerosis and attenuates inflammatory pain by regulating the expression of MMP9, MMP2, and CCL2.
