PD-L1 protein serves as a ligand for the inhibitory receptor PDCD1/PD-1 and is critical for maintaining immune tolerance. PD-L1 protein plays a crucial role in maintaining immune tolerance to self by acting as a ligand for the inhibitory receptor PDCD1/PD-1. This interaction modulates the activation threshold of T-cells, limiting their effector response and potentially stimulating T-cell subsets that produce interleukin-10 (IL10). However, tumors exploit the PDCD1-mediated inhibitory pathway to attenuate anti-tumor immunity and evade destruction by the immune system, thereby promoting tumor survival. The interaction between PD-L1 and PDCD1/PD-1 inhibits the function of cytotoxic T lymphocytes (CTLs), but blocking this pathway can reverse the exhausted T-cell phenotype and normalize the anti-tumor response, offering a promising strategy for cancer immunotherapy.
