Background

The PIGR protein serves as a crucial mediator in the selective transcytosis of polymeric IgA and IgM across mucosal epithelial cells. At the basolateral surface of epithelial cells, it binds to polymeric IgA and IgM, forming a complex that undergoes transportation across the cell to be ultimately secreted at the apical surface. Throughout this transcytosis process, a cleavage event separates the extracellular segment, known as the secretory component, from the transmembrane segment. PIGR, through its N-linked glycans, ensures the anchoring of secretory IgA (sIgA) molecules to the mucus lining the epithelial surface, contributing to the neutralization of extracellular pathogens. In its free form, PIGR may also act as a non-specific microbial scavenger, playing a role in preventing pathogen interaction with epithelial cells.

Biological Activity

1.Measured by its binding ability in a functional ELISA. When Recombinant Mouse plgR is coated at 2 μg/mL (100 μL/well), the concentration of mouse IgM that produces 50% of the optimal binding response is 297.7 ng/mL.
2.Measured by its binding ability in a functional ELISA . Immobilized recombinant mouse PIGR at 5 μg/mL (100 μl/well) can bind mouse IgM with a linear range of 0.156-10 μg/mL.

Reconstitution

Stored at -20°C for 2 years. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
