WEE2 Protein, Human (Sf9, His, GST)

Cat. No.: HY-P701729
COA Handling Instructions

WEE2 protein is an oocyte-specific tyrosine kinase that plays an important role in meiosis during prophase I and metaphase II. It maintains meiotic arrest at the germinal vesicle (GV) stage by phosphorylating CDK1 at 'Tyr-15', inhibiting CDK1 activity and preventing reentry. WEE2 Protein, Human (Sf9, His, GST) is the recombinant human-derived WEE2 protein, expressed by Sf9 insect cells , with N-8*His, N-GST labeled tag. The total length of WEE2 Protein, Human (Sf9, His, GST) is 566 a.a., .

Background

WEE2 Protein, an oocyte-specific protein tyrosine kinase, plays a pivotal role in regulating meiosis during both prophase I and metaphase II. Its essential function is highlighted in maintaining meiotic arrest in oocytes during the germinal vesicle (GV) stage, a prolonged quiescent period at dictyate prophase I. WEE2 achieves this by phosphorylating CDK1 at 'Tyr-15,' leading to the inhibition of CDK1 activity and preventing meiotic reentry. Furthermore, WEE2 is instrumental in facilitating metaphase II exit during egg activation by phosphorylating CDK1 at 'Tyr-15.' This action ensures the orderly exit from meiosis in oocytes, promoting pronuclear formation and contributing to the intricacies of reproductive processes.

Biological Activity

The activity of WEE2 is verified by the ADP-Glo method: Incubate WEE2 protein, ATP, and the substrate at room temperature for 40 minutes, then add the ADP-Glo reagent and read the luminescence signal using the chemiluminescence module of the microplate reader. The specific activity is 1327nM(IC50).

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
