    Semotiadil is a novel calcium antagonist whose stereochemical structure was investigated by X-ray crystallography and circular dichroism spectroscopy. X-ray structural analysis was performed using its enantiomeric salts with (S)-(+)-mandelic acid (2); 1 (mandelate salt). The R absolute configuration of 1, which was previously determined by chemical transformation of an optically active synthetic precursor, was directly confirmed by this analysis. Hydrogen bonding, charge repulsion, and hydrophobic interactions that stabilize the crystal structure were observed in the crystals. Circular dichroism spectra of 1 (mandelate salt) and 1 (hydrofuranate salt) in ethanolic solution showed similar spectral patterns. The CD bands of each chromophore were unambiguously assigned, and the conformation of the benzothiazole ring was determined using the optical rotation rule. The X-ray and CD data confirmed the P conformation of the benzothiazole ring and the equatorial position of the 2-phenyl ring in both the crystal and ethanolic solution states. These findings are in good agreement with our previous results on the conformational analysis of 2-arylbenzothiazole derivatives.

    4-Hydroxycanthin-6-one is a novel quinoline alkaloid isolated from the stem bark of the tree Ailanthus altissima. Five other known compounds were also found in the study. The structures of the new compounds were determined by interpretation of physical and spectroscopic data, and their absolute configurations were determined by electronic circular dichroism spectroscopy and quantum chemical calculations. These compounds showed significant inhibitory activity against lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW 264.7 cells, showing potential anti-inflammatory properties .

    NC-182 is a novel antitumor compound with a benzo[a]phenazine ring. The interaction mode of the agent with DNA and its effect on DNA conformation were systematically investigated by fluorescence, absorption and circular dichroism spectroscopy as well as viscosity titration measurements, and were compared with those of the DNA minor groove binder distamycin and the DNA base intercalator daunomycin. NC-182 was found to be a strong DNA intercalator, especially for B-form DNA, although without specificity for base pairs. The binding of NC-182 to B-DNA exhibited a biphasic behavior depending on the molar ratio of agent to DNA (r): at relatively low r values, NC-182 made the B-form structure rigid, and at higher r values, promoted the transition from B- to non-B-forms. NC-182 was also shown to promote the unwinding of Z-form DNA into B-form. Viscosity, UV 'melting' and circular dichroism experiments further showed that: (1) NC-182 intercalates to stabilize the DNA double helix structure; (2) NC-182 intercalates into the poly(dA) 2poly(dT) DNA structure to stabilize the triple helix structure, resulting in a melting point close to that of the double helix structure; the melting curves of the triple helix and double helix structures overlap at r > 0.06. These observations make an important contribution to our understanding of the biological properties of this novel benzo[a]phenazine derivative, which is a new antitumor agent suitable for both multiagent-resistant and sensitive tumors.

    BMVC4 is a G-quadruplex (G4) stabilizer of the human telomeric sequence d[AG3(T2AG3)3]. Screening by circular dichroism (CD) spectroscopy revealed that BMVC4 is more suitable as the core molecule of G4 stabilizers than BMVC. The results showed that BMVC4-12C and BMVC4-8C3O are better candidates for G4 stabilizers and are worthy of further study. A simple and rapid screening method based on Cu2+-induced G4 unfolding can be used to find better G4 stabilizers for potential anticancer applications. CD results showed that the trivalent cations of 9-substituted BMVC derivatives are more suitable as G4 stabilizers than the divalent cations of BMVC. In addition, by monitoring the disappearance of the 291 nm CD band of human telomeres after Cu2+ addition, it was found that the core molecule of G4 stabilizer BMVC4 has better stability.
