1. Membrane Transporter/Ion Channel
  2. Potassium Channel
  3. VU0546110

VU0546110 is a selective inhibitor of the sperm-specific potassium channel SLO3, with IC50s of 1.287 μM (SLO3) and 59.80 μM (SLO1) in HEK293 cells, respectively. VU0546110 blocks heterologous SLO3 currents and endogenous K+ currents in human sperm. VU0546110 halts sperm hyperpolarization, induced acrosome reaction, and hyper-activated motility. VU0546110 has contraceptive potential.

In Vitro

VU0546110 (10 μM) blocks the K+ current in human sperm at intracellular free Ca2+ concentrations of 0-50 µM[1].
VU0546110 (2.5 μM) inhibits the hyperpolarized membrane potential at pH 8.0 in human spermatozoa[1].
VU0546110 (2.5-10 μM) inhibits the hyperactivated motility and Ca2+ ionophore A23187 (HY-N6687) or progesterone (HY-N0437) induced acrosome reaction in human sperm[1].

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
