alpha-1,6-Fucosidase (LpAlfC(E274A)) (EC 3.2.1.51) cleaves branched non-reducing terminal fucose, linked α(1-6) to the core N-acetylglucosamine of N-linked oligosaccharides. alpha-1,6-Fucosidase (LpAlfC(E274A)) is useful for determining core fucosylation[1].

In Vitro

Product Information
A mutant of α 1,6-fucosidase, which can be used to attach fucose to GlcNAc-peptides, and is functionally reciprocal to LpAlfC (WT). This product is recombinantly expressed in E. coli, with a C-terminal His-tag, a molecular weight of 44 kDa, and a buffer composition of 20 mM Tris-HCl (pH 7.5), 10% glycerol.

Instructions
1. Add substrate according to the reaction system, add appropriate buffer salt to adjust the pH to 7.4, and add appropriate amount of enzyme to react (according to the reaction requirements);
2. After mixing, incubate the sample at 37℃ for 2-4 h or overnight.
Note:
1) When the system is scaled up, the incubation time or enzyme amount needs to be adjusted according to the actual situation, and the mixing can be done by low-speed shaking.
2) Most common biological buffers are applicable to the reaction system, such as Tris or PBS. Buffers that are not in this pH range (such as acetate buffer) may be applicable, but the incubation time or enzyme amount needs to be optimized according to the actual situation.
