  3. 2'-O-Methylcytidine

2'-O-Methylcytidine is an orally active 2'-substituted nucleoside as a inhibitor of HCV replication with antiviral activity. 2'-O-Methylcytidine inhibits RNA-dependent RNA polymerase (NS5B)-catalyzed RNA synthesis in vitro, in a manner that is competitive with substrate nucleoside triphosphate.

In Vitro

2'-O-Methylcytidine (0.01-100 μM, 24 h) inhibits HCV RNA replication with an IC50 value of 21 μM in HBI10A cells[1].
2'-O-Methylcytidine (2 μM, 23 h) yields very little triphosphate and suffers from extensive metabolism to species that are consistent with UTP and CTP in HBI10A cells[1].

In Vivo

2'-O-Methylcytidine (16.6 mg/kg, p.o., daily for 7 days) reduces the serum HCV RNA in chronically infected chimpanzees[3].

In Vivo:

For the following dissolution methods, please prepare the working solution directly. It is recommended to prepare fresh solutions and use them promptly within a short period of time.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
