9,10-Anthracenediyl-bis(methylene)dimalonic acid (ABMDMA) is a biological dye and indicator used to detect singlet oxygen generation (SOG). 9,10-Anthracenediyl-bis(methylene)dimalonic acid is water-soluble derivative of anthracene. 9,10-Anthracenediyl-bis(methylene)dimalonic acid can be photobleached by singlet oxygen to its corresponding endoperoxide. This reaction can be monitored spectrophotometrically by recording the decrease of absorbance at 400 nm[1][2].

In Vitro

Preparation of 9,10-Anthracenediyl-bis(methylene)dimalonic acid Staining Working Solution:[3]
0.15 mM of the sodium salt of 9,10-Anthracenediyl-bis(methylene)dimalonic acid in water was mixed with Pc4SNP or Pc4 to give a final concentration of 2 μM.
Detection of singlet oxygen generation:[1]
In a typical procedure, 0.5 mL of 9,10-Anthracenediyl-bis(methylene)dimalonic acid water solution (0.07 mg/mL) was added to methylene blue (MB)-GO or MB water solutions both containing 3.6 μg/mL (MB/H2O) to give the final 9,10-Anthracenediyl-bis(methylene)dimalonic acid concentration of 3.33 μg/mL. Next, 1 mL of MB-GO/9,10-Anthracenediyl-bis(methylene)dimalonic acid or MB/9,10-Anthracenediyl-bis(methylene)dimalonic acid solutions were poured into open quartz cuvettes and irradiated with a 785 nm laser for 55 min. The reactions were monitored with UV-vis spectroscopy by recording an absorbance decrease at 400 nm (λmax 9,10-Anthracenediyl-bis(methylene)dimalonic acid) at 5-min intervals.
