Description

Dextran (Dextran 40) has an inhibitory effect on thrombocyte aggregation and coagulation factors and is used as a plasma volume expander.

In Vivo

Dextran can be used in animal modeling to create paw edema models. Injection of dextran at doses of either 7.9 mg/kg or 300 mg/kg results in edema of equal intensity in the limbs[1][2].

Background
Dextran promotes the release of histamine and serotonin from mast cells during edema formation. These substances alter the tone and vascular permeability, contributing to fluid extravasation[1][2].
Specific Mmodeling Methods
Mice: Swiss albino• male • weighing 25-30 g[1].
Administration: 1% dextran (20 μL/paw)• intraplantar injection in the right hind paw• a single dose for 1, 2, 3 and 4 h[1].
Rat: Albino of of the Sprague Dawleq strain• weighing 110-130 g• seven-week-old[2].
Administration: 0.5 mL/100 g • tail vein injection• a single dose[2].

(1)Dextran (6% v/w in saline) was freshly diluted each day so as to contain 7.9 mg or 600 mg per 5 ml volume of physiological saline[2].
Modeling Indicators
Morphological change: Developed marked inflammatory reaction and significantly increased the volume of paw edema[1].
Histological analysis: Mast-cell disruption (histamine and serotonili release), vasodilatation, hypothermia, hypotension, helnoconcentration and edema occurred with the high doses of Dextran. Since minute doses of dextran induced only vasodilatation and edema formation[2].
