In Vitro

JNJ-70218902 binds to TMEFF2-positive LNCaP-AR cells (EC50: 9.6 nM) but not to TMEFF2-negative DU145 prostate cancer cells[1].
JNJ-70218902 has a high binding affinity for CD3 on primary human T cells (KD: 151 nM)[1].
JNJ-70218902 induces T cell-mediated killing (EC50: 1.4 nM) when incubates with healthy human donor T cells and LNCaP-AR cells at an effector-target ratio of 3:1, and T cell activation is demonstrated by increased cell surface CD8+CD25+ expression and pro-inflammatory cytokines[1].
JNJ-70218902 shows low cytotoxicity in DU145 cells[1].

In Vivo

JNJ-70218902 has antitumor activity at increasing concentrations up to 5 mg/kg in T cell humanized NSG mice bearing LNCaP xenografts (tumor growth inhibition rates of 75-122%) and LuCaP 86.2 patient-derived xenografts (tumor growth inhibition rates of 72-88%)[1].
JNJ-70218902 (0.075 mg/kg or 0.075 mg/kg, then 0.3 mg/kg after one week) in a cynomolgus monkey model of prostate cancer can induce the infiltration of activated and proliferating T cells and inflammatory cells into the prostate, mediating an effective immune response[2].
