Cutinase Protein, Thermobifida Fusca (His) is the recombinant Cutinase protein, expressed by E. coli , with C-6*His labeled tag. The total length of Cutinase Protein, Thermobifida Fusca (His) is 261 a.a., with molecular weight of 28-30 kDa.

Background

The Cutinase protein assumes a pivotal role in cellular processes, catalyzing the hydrolysis of cutin, a key polyester comprising the plant cuticle structure. As documented in various studies, Cutinase exhibits esterase activity toward p-nitrophenol-linked aliphatic esters (pNP-aliphatic esters), displaying a preference for short-chain substrates. Moreover, Cutinase demonstrates versatility by hydrolyzing triglycerides like triacetin, tributyrin, tricaprin, and trilaurin, with a pronounced affinity for short-chain substrates. Additionally, this enzyme displays the ability to hydrolyze the hemicellulose xylan and exhibits proficiency in degrading the widely used plastic poly(ethylene terephthalate) (PET). Furthermore, Cutinase can depolymerize poly(epsilon-caprolactone) (PCL), a synthetic aliphatic biodegradable polyester, highlighting its role in various biodegradation processes. The broad substrate specificity of Cutinase underscores its significance in enzymatic reactions involved in natural polymer breakdown and synthetic polymer degradation, showcasing its potential applications in environmental and industrial contexts.

Biological Activity

Measured by its ability to catalyze a substrate, p-nitrophenol succinate, which can be measured by absorbance at 410 nm. The specific activity is 42.67 U/mL, as measured under the described conditions.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
