Background

The DKK-3 protein functions as an antagonist of canonical Wnt signaling by impeding the interaction between LRP5/6 and Wnt and forming a ternary complex with the transmembrane protein KREMEN, facilitating the internalization of LRP5/6. Within vertebrate development, DKKs, including DKK-3, play a crucial role by locally inhibiting Wnt-regulated processes such as antero-posterior axial patterning, limb development, somitogenesis, and eye formation.
