Description

DKK-3 protein antagonizes canonical Wnt signaling by inhibiting LRP5/6-Wnt interaction. It forms a ternary complex with KREMEN and promotes LRP5/6 internalization. DKK-3 Protein, Mouse (HEK293, His) is the recombinant mouse-derived DKK-3 protein, expressed by HEK293 , with C-His labeled tag.

Background

The DKK-3 protein is involved in antagonizing canonical Wnt signaling by inhibiting the interaction between LRP5/6 and Wnt. It achieves this through the formation of a ternary complex with the transmembrane protein KREMEN, which facilitates the internalization of LRP5/6. DKKs, including DKK-3, are crucial in vertebrate development as they locally inhibit Wnt-regulated processes such as antero-posterior axial patterning, limb development, somitogenesis, and eye formation. In adult organisms, DKKs are implicated in bone formation, bone diseases, cancer, and Alzheimer's disease. Additionally, DKK-3 interacts with LRP5 and LRP6 proteins. It is important to note that DKK-3 does not possess carboxypeptidase activity or dipeptidyl-peptidase IV type activity.

Biological Activity

Measured by its ability to inhibit proliferation of HeLa human cervical epithelial carcinoma cells. The ED50 for this effect is 4.628 μg/mL, corresponding to a specific activity is 216.076 units/mg.
