Background

Frizzled-8, functioning as a receptor for Wnt proteins, plays a pivotal role in the intricate Wnt-Fzd-LRP5-LRP6 complex, orchestrating beta-catenin signaling by inducing the aggregation of receptor-ligand complexes into signalosomes of ribosome size. Operating primarily through the canonical Wnt/beta-catenin signaling pathway, it prompts the activation of disheveled proteins, inhibits GSK-3 kinase, facilitates nuclear accumulation of beta-catenin, and activates Wnt target genes. An additional signaling pathway involving PKC and calcium fluxes has been observed in some family members, although the extent of its integration with the canonical pathway remains unclear.
